中药调控PI3K/AKT信号通路干预黑色素瘤的研究进展

摘要:恶性黑色素瘤(Malignant melanoma,MM)是一种源于黑色素细胞异常生长的恶性肿瘤,有高转移性和高致死性,后者占皮肤癌相关死亡比例>80%。PI3K/AKT信号通路的异常激活会促进MM的发生,在MM病理进程中具有重要意义。目前研究发现,多种中药对恶性黑色素瘤具有很好的效果。本文基于PI3K/AKT信号通路系统总结中药复方及注射液和单体干预MM的作用机制,发现中药复方及注射液(归芪白术方、复方苦参注射液、四君子汤)和中药单体萜类(人参皂甙、三七总苷、α-常春藤皂苷等)、酚类(白藜芦醇、姜黄素、山萘酚等)、黄酮类(半枝莲总黄酮、新藤黄酸、淫羊藿苷等)等均可通过抑制PI3K/AKT信号通路的关键分子,从而抑制MM。基于此,本文总结了中药单体及复方干预PI3K/AKT信号通路干预黑色素瘤的研究进展,以期为中药防治恶性黑色素瘤的临床应用及新药研发提供思路。

Research progress of traditional Chinese medicine in regulatingPI3K/AKT signaling pathway to interfere with malignant melanoma

Abstract:Malignant melanoma (MM) is a kind of malignant tumour originating from the abnormal growth of melanocytes, It has a high degree of metastasis and lethality, with the latter accounting for more than 80% of skin cancer-related deaths. Abnormal activation of PI3K/AKT signaling pathway can promote the occurrence of MM,which has important significance in the pathological process of MM. Current research has found that A variety of traditional Chinese medicine has a good effect on malignant melanoma.In this paper, we summarise the mechanism of action of Chinese herbal compounds and monomers intervening in MM based on the PI3K/AKT signalling pathway, and find that Chinese herbal compounds (Guiqi Baijutsu Formula, Compound Bitter Ginseng Injection, Sijunzi Tang) as well as Chinese herbal monomers of terpenoids(ginsenoside, total Panax ginseng glycosides, α-Ivy saponins, etc.),phenolics(resveratrol,curcumin,sennosides,etc.),flavonoids (hemicarpene total flavonoids, neo garcinia cambogia acid, epimedium glycosides, etc.) etc. can all reduce MM by affecting the PI3K/AKT signalling pathway. Based on this, this paper summarized the research progress of TCM monomer and compound intervention of PI3K/AKT signaling pathway in the intervention of melanoma, in order to provide ideas for the clinical application of TCM in the prevention and treatment of malignant melanoma and the development of new drugs.
